No, the update works over-the-air (at least with wifi), but it is a two-stage-process on the iphone 5 which had the bug. Opening settings/software-update loads a fixed updateprogram which when started will load the real 6.0.1-update.

I updated mine using WiFi, so it may only be specific circumstances that require connecting directly to computer.
I upgraded to 6.0.1 as soon as I noticed that it was available. I did it over 3G from my office, which has a weak signal, and it was done in less than 5 minutes, including the reboot.

The iOS 6.0.2 update is available only to iOS developers right now. 6.0.1 is the latest, and it actually made the problem worse on my 4S. Prior to the update, I could reboot my 4S and Waze would load and work for a while. After updating to 6.0.1, it launches and quits immediately no matter what. Come on Waze, quit wasting time on the Android app and fix the one with all the marketshare!

iOS 6.0.1 can be loaded over the air now, at least on iPhone 5 via AT&T. It downloads an app to your phone that then works around the bug to update iOS to 6.0.1 and conveniently removes itself during the update process. A rather elegant solution, I must say...
